Abstract: | The thesis contains the results of an investigation on the " Population Genetic Structure of the Penaeus indicus " from southeast and southwest coasts of India. The P.indicus, popularly known as the Indian white prawn, is distributed widely in the Indo-Pacific, starting from New South wales in Australia in the east to the east coast of Africa in the west. Its heavy demand in the export market, the species has been exploited intensively from all along its areas of distribution in Indian waters. The population genetic characteristics of the species were examined by three independent but complementary techniques, namely, morphometrics (truss network), biochemical genetics (isozyme electrophoresis ) and molecular genetics (RFLP and RAPD). The east and west coast populations of the species may be genetically different. Due to certain constraints, the results obtained from the studies of restriction fragment length 70 polymorphism (RFLP) were limited. The significant difference in the number of bands in the sample populations strongly suggests that these two populations have considerably different population genetic structures |

Abstract: | The work presented in this thesis is centered mainly around delineating the toxic effect of hydrogen sulphide on penaeid prawns and understand its influence on the ecology of estuary. The present investigation also involved characterization of the effects of hydrogen sulphide on the growth and behavioural responses of Penaeus indicus. The test animals employed during the present study namely indicus and Metapenaeus dobsoni are both ecologically and economically relevant. The thesis embodying the details of the investigation has been organized into three chapters comprising Acute toxicity, influence of hydrogen sulphide on the ecology of estuary and effect of hydrogen sulphide on growth and substratum selectivity of penaeid prawn. Each chapter has been partioned into various sections as Introduction, Material and Methods, Results and Discussion for a lucid presentation of the subject matter. |

Abstract: | The objective of the study isto determine the average quantity of certain biochemical constituents of the haemolymph of Penaeus indicus and to verify the importanceof the simple correlation between the quantity or content of the biochemical constituents in the haemolymph and the size of the species, sex, moult and reproductive stages. The biochemical constituents studied are protein, free amino acids, glucose, total lipids, cholesterol, calcium, zinc, iron and manganese. The study Identifies the species specific haemolymph protein pattern by electrophoresis and determines the qualitative variations of haemolymph proteins with respect to sex, size, moult and reproductive stages. Major protein components such as hemocyanin and female specific protein are determined with a view to understand their function. The thesis also identifies the circulating haemocytes with a view to understand their specific role in the various physiological functions of the species. The thesis is presented in three chapters. Each chapter has an introduction to the particular aspect of study which includes a review of literature, methodology adopted for the study, the results obtained and discussion on the subject. The first Chapter deals with the biochemical constituents of the haemolymph, the second includes electrophoretic characterization of proteins in the haemolymph and the third Chapter deals with haemocyte identification and classification. A summary of the thesis and literature cited in the text are listed at the end. |

Abstract: | The aim of the present investigation is to build up the knowledge on the role of commensal bacteria present on the prawns during storage at various temperatures. The study Evaluates the nature of spoilage of prawns during storage at three different temperatures (28:2OC, 4°C and -18°C) by organoleptic assessment, accumulation of trim ethylamine, ammonia content, changes in the flesh pH and total heterotrophic bacterial population at various time intervals and to find out the changes in the proximate composition (protein, carbohydrate, lipid, ash and moisture) of the prawns during storage at various temperatures by estimating the contents at different time intervals along with spoilage assessment. The researcher studies the occurrence and role of various bacterial genera which form the component of spoilage flora during storage and determines the distribution of various hydrolytic enzyme producing bacteria by evaluating their ability to produce enzymes such as caseinase, gelatinase, amylase, lipase and urease. to assess the spoilage potential of the bacteria by testing their ability to reduce trimethylamine oxide (TMAO) to trimethylamine (TMA) and to produce odour in flesh broth and halos in flesh agar media.The researcher also gives stress on the growth kinetics of selected potential spoilers by growing_them in different media and to assess the effect of sodium chloride concentrations, temperature and pH on their growth, survival and. generation time. |

Abstract: | The thesis presents the results of the studies carried out on certain diseases encountered in the larvae and postlarvae of penaeid prawns raised in the hatcheries at Cochin, Madras and Mandapam Camp during September 1985- April 1988. In the preliminary survey carried out to understand the common diseases occurring in the penaeid larvae and postlarvae, seven cases of diseases and abnormalities were encountered. These included ciliate infestation, Nit_zschia closteriurn infestation, parasitic protozoaninfection, parasitic dinoflagellate infection, appendage necrosis, heteromorphic eye and abnormal eggs and deformed nauplii .The clinical signs, seasonal occurrence and incidence of each of the above cases were provided along with the information on environmental factors such as salinity, dissolved oxygen, temperature and pH of the rearing medium. The Thésis is presented in nine chapters. Chapter 1 surveys the literature on the diseases of penaeid larvae, postlarvae and adult prawns from India and abroad. This is followed by a chapter on the material and methods employed during the present investigation. In the third chapter, seven cases of diseases and abnormalities encountered in the larvae and postlarvae of Penaeus indicus and p. semisulcatus during the survey carried out in the hatcheries located at different centres of Central‘ Marine Fisheries Research Institute are presented and discussed .A bacterium responsible for appendage necrosis was isolated and its taxonomy was studied. It was Gram-negative, fermentative and motile rod. It was sensitive to vibriostatic compound, 0/129. This bacterium was found to be a new isolate of vibrio on the basis of its morphological, biological, physiological and biochemical characters and comparison of these characters with those described for other related vibrios. This new isolate of vibrio was deposited in vibro Referrence Laboratory, Centres for Disease Control, Georgia, U.S.A. and coded as vibrio sp. 2448-88. |

Abstract: | The present investigation has been carried out to understand the process and events leading to maturation of the ovary and testis in the Indian white prawn Penaeus indicus. The study includes the classification of the ovarian maturity stages based on its colour, gonadosomatic index, oocyte diameter and morphological changes in the oocyte. Further the process of oogenesis has been investigated using light and electron microscopic techniques. A histochemical study of the ovary has also been carried out to determine the sequence in which yolk substances are synthesized or sequestered in the oocytes and also to elicit the nature of the penaeid yolk material. The process of spermatogenesis and the development of the spermatophore has been studied in detail using light and electron microscopic methods. In addition a brief histochemical study on the testis was also made to understand the nature of the organic reserves in the sperm cells. |

Abstract: | The present study is the first comprehensive approach towards histopathology of White Spot Syndrome Virus (WSSV) in Penaeus indicus. WSSV could be demonstrated in the nuclei of all tissues, except those of midgut, subjected of electron microscopic observation. They were the nuclei of gill, foregut, heart, hepatopancreatic connective tissue, hindgut, nerve and dorsal aorta. A comparison was made between the electron microscopic and histopathological observations and a greater degree of correlation between the two in depicting the severity of the infection of the infection was unraveled. The study also illustrated variations in response and susceptibility of various tissues to WSSV infection. Accordingly, out of the tissues investigated, gill, foregut, hindgut and dorsal aorta exhibited advanced viral multiplication than the other tissues such as heart, midgut, nerve and hepatopancreas. Even though hepatocytes were not infected the connective tissue nuclei were packed with virions. |
