Abstract: | Vibrio sp. V26 isolated from mangrove sediment showed 98 % similarity to 16S rRNA gene of Vibrio cholerae, V. mimicus, V. albensis and uncultured clones of Vibrio. Phenotypically also it resembled both V. cholerae and V. mimicus.Serogrouping, virulence associated gene profiling, hydrophobicity, and adherence pattern clearly pointed towards the non—toxigenic nature of Vibrio sp. V26. Purification and characterization of the enzyme revealed that it was moderately thermoactive, nonhemagglutinating alkaline metalloprotease with a molecular mass of 32 kDa. The application of alkaline protease from Vibrio sp. V26 (APV26) in sub culturing cell lines (HEp-2, HeLa and RTG-2) and dissociation of animal tissue (chick embryo) for primary cell culture were investigated. The time required for dissociation of cells as well as the viable cell yield obtained by while administeringAPV26 and trypsin were compared. Investigations revealed that the alkaline protease of Vibrio sp. V26 has the potential to be used in animal cell culture for subculturing cell lines and dissociation of animal tissue for the development of primary cell cultures, which has not been reported earlier among metalloproteases of Vibrios. |

Abstract: | A total of 34 yeast isolates were characterized from 4 water samples collected from Kongsfjord at Ny Alseund region of Norwegion Artic during the Indian Artic summer expedition of 2009.They were studied for the effect of tempereture and salt concentration on growth as well as for their ability to produce various hydrolytic enzymes at two different temperatures. Result showed that 5 out of 8 genera were common to all the stations. Cryptococcus was the predominant genera folowed by Trichosporan and Rhodotorula 82% of the yeast isolates were oxidative in nature and except filobasidium all the isolates used nitrate as a nitrogen source for growth. Yeast isolates from all the ststions showed growth at 4 and 20 degree centigarade. These temperatures were chosen as most of the bacterial and yeast isolates showed psychrotrop[hic nature. 94% of the yeast isolates showed growth at 2.0M and lipolytic activity were marginally less than 4.None of the isolates produced amylase enzymes when incubated at 4 and 20. The present study highlights the wide tolerence of the psychrotrophic yeast isolates to temperature and salinity as well as their potential in biotechnology |

Abstract: | Emergence of antibiotic resistance among aquaculture pathogens has made it necessary to look into environment friendly, effective and sustainable methods such as probiotic and immunostimulants among others.. In the present study, LAB were isolated from the gut of fish species namely Rastrelliger kanagurta and analyzed for their antibacterial activity against various fish, shrimp and human pathogens. Different LAB species such as Lactobacillus plantarum, L. bulgaricus, L. brevis and L. viridiscens were encountered in the gut of R. kanagurta. Several strains showed good activity against fish, shrimp and human pathogens. LAB from the gut of such marine species may be developed as possible probiont for environment friendly health management of fresh water, estuarine and marine species currently exploited in aquaculture |

Abstract: | The main objectives of the present investigation were to evaluate the qualitative and quantitative distribution of natural cyanobacterial population and their ecobiological properties along the Cochin estuary and their application in aquaculture systems as a nutritional supplement due to their nutrient-rich biochemical composition and antioxidant potential. This thesis presents a detailed account of the distribution of cyanobacteria in Cochin estuary, an assessment of physico-chemical parameters and the nutrients of the study site, an evaluation of the effect of physico-chemical parameters on cyanobacterial distribution and abundance, isolation, identification and culturing of cyanobacteria, the biochemical composition an productivity of cyanobacteria, and an evaluation of the potential of the selected cyanobacteria as antioxidants against ethanol induced lipid peroxidation. The pH, salinity and nutritional requirements were optimized for low-cost production of the selected cyanobacterial strains. The present study provides an insight into the distribution, abundance, diversity and ecology of cyanobacteria of Cochin estuary. From the results, it is evident that the ecological conditions of Cochin estuary support a rich cyanobacterial growth. |

Abstract: | Considering the extent of warming in the artic region and the resultant changes in the dynamic marine enviornments there is a need to monitor the bacterial diversity in the fjord enviornments especially in terms of cultivable bacteria. The present study reports the diversity of cultivable hetrotrophic bacteria from the water and sediment samples of kongsfjord their growth responses to important enviornmental variables and ability to produce industrially important hydrolytic enzymes. |

Abstract: | This study aims to reveal the ability of bacteria isolated from Cochin estuary and the Arabian Sea to produce phosphatases, important characters of the enzymes, its ability to utilize organophosphorus compounds as source of phosphate and also their role in degradation of organophosphorus pesticides. It deals with isolation, identification and screening of bacteria for phosphatase production, and it describes the effect of cultural conditions on growth and phosphatase production. The effect of various factors like pH, NaCl concentration, temperature of incubation, carbon source, period of incubation etc. on growth and phosphatase production by the two selected species were studied to establish suitable environment for phosphatase production by these bacteria. In this study regulation of phosphatase synthesis, characteristics of acid and alkaline phosphatases are discussed. |

Abstract: | This thesis Entitled Studies on amylolytic bacteria in cochin backwaters.This thesis presents a detailed account of the disribution of amylolytic bacteria in water. sediment. fishes ( Etroplus suratensis and Liza parsia) • prawns ( Penaeus indicus and Metapenaeus dobsoni) and clams ( Sunetta scripta and Meretrix casta) from Cochin backwaters. genera-wise distribution of amylolytic bacteria, ability of selected strains to grow and produce amylase at various physico-chemical conditions. Regulation of amylase synthesis anrt characters of amylases producer by these halophilic bacteria.Amylolytic bacteria are distributed widely in water. sediment. fishes. prawns and clams of Cochin back waters. 53% of the total isolates tested were capable of producing amylase. Maximum number of arnylolytic bacteria were present in Metapenaeus dobsoni. In general, the gut region of aquatic animals harboured more amylolytic bacteria than the gill or surface. These bacteria may help in the digestion of starch present in their food.Presence of ions in the medium was found to be essential for growth and amylase production. It was found that this ionic requirement is not highly specific. Sorlium chloride could be replaced by potassium chloride. or magnesium chloride to some extent I without affecting growth and amylase production. The important function of these ions may be to maintain the osmotic balance between the cells and their environment.All the isolates showed the ability to grow and produce amylase using raw-starches from cassava. plantain and potato .This property suggests their role in the rdegradation of native starches in the environment |
